Acute oral toxicity study on rats

1. Test purpose:
determine the LD50 according to the test results; assess primarily poisoning targets and potential toxicity action mechanism; provide reference to dosage level design of subchronic, chronic and other toxicity test; provide criterion for acute toxicity classification and safeguard¡¯s establishment.

2. Materials and method:

2.1 test pesticide: Cymoxanil tech, offwhite powder, was provided by Saturn Agrochemical Inc.

2.2 test animals: 40 SD rats with weight 180~220g (female and male rats 20, respectively) were provided by Nanjing Medical University Animals Center. All the animals were fasted whole night prior to administration. Cymoxanil was diluted to different concentration with salad oil. Then the test pesticide was administrated orally to animals by stomach for one time according to 10ml/kg b.w.. Environment temperature of feeding animals was 20¡À2¡æ, humidity was 50%-80% and it was well ventilated. Weigh every 3d during whole observation period.

2.3 test method: all rats were divided randomly into 4 groups with each 5 rats/sex. Each group was fed Cymoxanil solution at the doses of 1000, 2150, 4640 and 10000mg/kg, respectively at room temperature. Feed after treatment for 2h. Observed poisoning symptom and mortality circumstance for two weeks after treatment to calculate LD50 according to Horn method.

3 The results:
After administration, animals showed some poisoning symptoms such as reducing action, cachexia etc. Death was observed within 72h. The survival recovered gradually 4d later. After dissection, no macroscopic pathological changes could ben found in every essential organ. The test showed that LD50 for female rats was 2000mg/kg and for male rat was 2710mg/kg. Cymoxanil was classified as low toxic pesticide.
